5-Amino-1MQ (5-amino-1-methylquinolinium) is a small-molecule research compound — not a peptide — developed as a selective, membrane-permeable inhibitor of nicotinamide N-methyltransferase (NNMT). NNMT is a cytosolic enzyme that transfers a methyl group from S-adenosylmethionine (SAM) to nicotinamide, producing 1-methylnicotinamide.

Because nicotinamide is a precursor in the NAD+ salvage pathway and SAM is the cell's main methyl donor, NNMT sits at the junction of NAD+ metabolism and cellular methylation. NNMT expression is elevated in the adipose tissue of obesity models, which made the enzyme a target for metabolic research.

In published work from the University of Texas Medical Branch (Neelakantan et al., Biochemical Pharmacology, 2018), 5-amino-1MQ lowered 1-methylnicotinamide and raised intracellular NAD+ and SAM levels in cultured adipocytes, and treating diet-induced obese mice with the compound was associated with lower body weight and white adipose tissue mass. A later study by the same group (2019) examined NNMT inhibition with 5-amino-1MQ in mouse models of aged skeletal-muscle regeneration. These are preclinical findings in animal and cell models; they have not been demonstrated in humans.
