A Certificate of Analysis (COA) is the most important quality document accompanying any research peptide. It provides independent laboratory verification of the identity, purity, and quality of the compound. Understanding how to read and verify a COA is essential for any researcher working with peptides. What is a COA?
A Certificate of Analysis is a document issued by an independent laboratory that certifies test results for a specific batch. For research peptides, it typically includes:
- HPLC purity analysis — measures the percentage of target peptide, typically ≥98% for research grade
- Mass spectrometry (MS) — confirms molecular identity by measuring molecular weight
- Amino acid sequence verification — confirms the correct sequence
- Appearance and solubility — physical characteristics of the lyophilized powder
- Batch/lot number — unique identifier for the production batch
Step-by-Step Verification
- Locate the COA. Every Pepspan order includes a COA. If you cannot find yours, contact [email protected].
- Check the testing laboratory. Look for an independent, accredited lab (not the manufacturer). Pepspan uses Janoshik Analytical for third-party verification.
- Verify peptide identity via mass spectrometry. The observed molecular weight should match the theoretical MW within ±1 Da. For example, BPC-157 has a theoretical MW of 1419.53 Da.
- Review HPLC purity results. Look for a single dominant peak with purity ≥98%. Multiple large peaks may indicate impurities.
- Match the batch number. The batch number on the COA must match your vial label exactly.
- Check the testing date. Results should be recent and from the current production batch.
Red Flags to Watch For
- No laboratory name or accreditation information
- Purity below 95% for research-grade peptides
- Missing mass spectrometry data
- Batch number does not match the vial
- Generic or template COA without specific results
- COA issued by the manufacturer (not independent)
